Effect of Calcitonin, NaF and Tamoxifen on Osteoporosis Induced by Ovariectomy in Rat



Abstract
The effects of calcitonin, Naf and Tamoxifen on osteoporosis were evaluated after inducing osteoporosis in the rat by overiectomy. One hundred rats were ovariectomized at the beginning of the study and among these, twenty rats were sham-operated
and
eighty rats were ovariectomized. Sham-operated twenty rats(group 1) had no injection. In group 2(twenty rats), 0.5cc of normal saline was injected intramuscularly 6 times a week for 16 weeks. In group 3(twenty rats), 2.5 MRC unit/kg of
(Asu1.7)-eel
calcitonin was injected intramuscularly 6 times a week for 16 weeks. In group 4(twenty rats), these were fed with 1 mg/kg of NaF 6 times a week for 16 weeks. In group 5(twenty rats) 0.2mg/kg of tamoxifen was given 6 times a week for 16 weeks. All
rats
were killed with suffocation at the 16th week. Sections of the femoral shafts were stained with Villanueva bone stain and evaluated histomorphometrially. Ovariectomy produced a decreased weight of the femur and significant increases of the
medullary
area. Periosteal bone formation did occur. The increase of the medullary area resulted from an increase of the number and size of osteoclasts.
In contrast, treatment of the ovariectomized rats with calcitonin, NaF and tamoxifen for 6 weeks prevented the increase of the medullary area and the decrease of the trabecular area in ovariectomized rats. Also the activity of osteoclast was
inhibited
by these treatments. The new bone formation rate was the highest in the NaF treated group and followed by calcitonin and tamoxifen treated groups. These results indicated that, in the ovariectomized rats, bone resorption was inhibited the most in
the
NaF treated group, and calcitonin and tamoxifen had less effects on the inhibition of bone resorption compared with NaF.
